The electricity-powered, air-conditioned buses made their debut on Sunday, with fares starting at 40 baht and ending at 120 baht.
The 51-kilometre Route 8580 offers 16 trips from 5.45am to 8pm, at roughly one-hour intervals, with 25 stops along the way.
The new line, the fifth on the island, is operated by the Phuket Provincial Administration Organisation.
The office plans to extend the route from the airport to the Tha Chatchai checkpoint on the northern tip of the island, Phuket PAO president Raywat Arreerob said on Tuesday.
